Looking at the Defaults.py on my MM 2.1a3 test installation I notice that 
it reads as follows:

# Site-specific settings
DEFAULT_EMAIL_HOST = 'mymachine.mydomain.co.uk'
DEFAULT_URL_HOST = 'mymachine.mydomain.co.uk'
DEFAULT_URL_PATTERN = 'http://%s/mailman/'

# For backwards compatibility.  Note: DEFAULT_URL_PATTERN must end in a slash!
DEFAULT_HOST_NAME = DEFAULT_EMAIL_HOST
DEFAULT_URL       = DEFAULT_URL_PATTERN % DEFAULT_URL_HOST

Adding a definition for DEFAULT_EMAIL_HOST='mysite.net' to your mm_cfg.py 
may be worthwhile.

If you are creating your new lists through the web rather than with 
bin/newlist I think you also need a call to add_virtualhost (see comments 
in Defaults.py) or redefine VIRTUAL_HOSTS = {} in mm_cfg.py
