I recently announced an article describing a pure Python system
(http://www.amk.ca/python/writing/mx-architecture/), though I wouldn't
call the system or database large, as such things go.  

The pieces for assembling Python-based business applications are all
there -- ZODB, relational database support, Web application servers --
but they're not packaged or documented together.  It would make an
interesting project for someone.
