As far as GUI builders go, Glade (PyGTK) and wxDesigner (wxPython) are
really nice.  Glade+libglade is a really cool way to build a GUI.
I've just started to try out wxDesigner, and it seems to be a little
odd in the way it organizes the code it generates.  It might just be
that I'm not used to it ...
