False.

Hostnames are explicitly case insensitive.  Mailman flattens the
strings prior to comparison.  Example:

  https://www.kanga.nu/lists/listinfo/
  https://WwW.Kanga.Nu/lists/listinfo/

Both will report the same page.  As for the setting:

 $ fgrep VIRTUAL_HOST_OVERVIEW /var/lib/mailman/Mailman/*.py
 /var/lib/mailman/Mailman/Defaults.py:VIRTUAL_HOST_OVERVIEW = 1
