¿Have you updated DNS? Something similar happened to me.
In my organization updated DNS to a new version, so asking localhost in
DNS doesn't resolve now.

I had in my Defaults.py:
SMTPHOST          = 'localhost'
because I have in the same machine mailing lists and mail server.

I changed
SMTPHOST = 'canonical.name.of.smtp.server'
and then it worked!, it was very hard to find the bug :-(

Try it and tell if it has worked
